چکیده

tion organelle. Primary cilia, immotile organelles found on most cells, have been implicated in several signaling pathways and we have observed direct binding of Hh protein to them. We find that the Ptc receptor protein is localized in cilia, where it prevents the accumulation of the Smo seven-transmembrane domain protein. Binding of Hh to Ptc causes departure of both from the cilium, allowing entrance of Smo. Smo in the cilium is able to activate Gli transcription factors which in turn control target gene expression. We have identified oxysterols as powerful agonists of Hh signaling in mammalian cells, and find that they also cause Smo to move into cilia. Using agonists and antagonists of Hh signaling, and mutants that affect primary cilia, we are exploring the mechanisms of protein trafficking and target gene activation. We are also characterizing Hh target genes in responsive cerebellum granule neuron precursors and in the medulloblastoma tumors that arise from these precursors when Ptc function is reduced.

Analysis of epithelial mesenchymal transition markers in breast cancer cells in response to stromal cell-derived factor 1

Introduction: Metastasis is the main cause of cancer death; however, the underlying mechanisms of metastasis are largely unknown. The chemokine of stromal cell-derived factor 1 (SDF1) and the process of epithelial mesenchymal transition (EMT), both have been declared as important factors to promote cancer metastasis; however, Conspicuously, the relation between them has not been recognized well...

Modeling tumor invasion and metastasis in Drosophila

Conservation of major signaling pathways between humans and flies has made Drosophila a useful model organism for cancer research. Our understanding of the mechanisms regulating cell growth, differentiation and development has been considerably advanced by studies in Drosophila. Several recent high profile studies have examined the processes constraining the metastatic growth of tumor cells in ...

Microwave Tumor Ablation of Vertebral Bone Metastasis: A Case Report

Introduction: Thermal ablation by microwave (MWA) has proven efficacy in treatment of primary and metastatic cancers. Soft tissue tumors could be ablated completely using MWA. However, bony tissue metastasis particularly spinal vertebral metastasis ablation has not reported yet. Case presentation: A 50 year old male patient with primary diagnosed colon cancer undergone surgical resection of ...

Breast Cancer Metastasis to Pituitary Infandibulum

Metastasis from breast cancer to other parts of the body is very common, but the spread of the tumor to pituitary gland, especially to infandibulum, is a rare presentation. At the time of pituitary metastasis, a majority of the patients have clinical and radiological evidence of the disease. It seems that the posterior area of the gland is the most common site of metastasis, probably due to hig...

Molecular targets of pomegranate (Punica granatum) in preventing cancer metastasis

Metastasis is the primary cause of mortality and morbidity among cancer patients and accounts for about 90% of cancer deaths. The most common types of treatment for cancer metastasis are chemotherapy and radiotherapy. However, such therapy has many serious side effects that could diminish the quality of life in patients. There is increased appreciation by the scientific community that natural c...
